Rapid Strep A Test Kit (Throat Swab Dipstick)

A Rapid Strep A test kit for throat swabs is a point-of-care diagnostic kit used to quickly detect Group A Streptococcus (Strep A) bacteria – the cause of “strep throat” – in a patient’s throat specimen. These kits use a lateral-flow (immunochromatographic) dipstick or cassette device coated with antibodies that bind Streptococcus group A antigen. In practice, a sterile swab is rubbed on the patient’s tonsils/pharynx and then mixed with an extraction buffer. The dipstick (or test cassette) is inserted into the sample mixture, and within about 5 minutes a colored line appears if Strep A antigen is present. A second “control” line should always appear to confirm the test ran correctly. For example, Quidel’s QuickVue® Dipstick Strep A test “detects group A Streptococcal antigen directly from patient throat swab specimens” and is used as “an aid in the diagnosis of group A Streptococcal infection”. Similarly, other manufacturers describe their Strep A rapid kits as “rapid chromatographic immunoassays” for qualitatively detecting Strep A antigen from throat swabs.

The Rapid Strep A Testing Kit Throat Swab Specimen Dipsticks are utilised for the quick diagnosis of strep throat, caused by the Streptococcus pyogenes bacterium. This essential point-of-care test enables healthcare providers to swiftly determine the presence of a bacterial infection, differentiating it from viral infections. It involves using a throat swab to collect a sample, and the dipsticks deliver accurate results in minutes. Prompt diagnosis allows for timely antibiotic treatment, reducing the risk of complications and limiting the spread of the infection.

How the Test Works

The core of the kit is a nitrocellulose test strip (dipstick) with two key antibody regions. One region (the test line) is coated with anti–Strep A antibodies; the other is a control line with antibodies that bind a built‐in control. When the swab sample (mixed in buffer) is applied, any Strep A antigens in the sample bind colloidal gold (or colored) particles conjugated to antibodies. Capillary action pulls the mixture along the strip. If Group A strep antigen is present, it will be captured at the test line and form a visible colored band. A colored control line will always form downstream to indicate the test strip is working properly. In short, two visible lines = positive (Strep A detected); one line (control only) = negative. These devices are purely qualitative – they indicate presence or absence of antigen, not amount.

The test reagents (swabs, buffers, dipsticks) are usually supplied in a self-contained kit. A typical kit (often sold in boxes of 20–25 tests) includes sterile throat swabs, pre-measured buffer tubes (extraction solution), and individually sealed dipstick cassettes or strips. For example, one commercial kit contains 20 individually wrapped test cassettes, 20 dropper bottles of extraction buffer (commonly a nitrite/acetic acid solution), and 20 sterile swabs. The testing process is straightforward:

  • Collect sample: Using the provided sterile swab, rub the back of the patient’s throat/tonsils (avoiding the tongue) to collect mucous and epithelial cells.
  • Extract antigen: Place the swab into the extraction tube containing buffer. Some kits use a one- or two-step chemical buffer (e.g. sodium nitrite and acetic acid) to break open cells and release antigen. Shake or swirl the swab in the solution for about 2–5 minutes so the antigen diffuses into the liquid.
  • Apply to test: Remove the swab, squeezing it against the tube wall to recover liquid. Then either dip the test strip (dipstick) into the tube or add a dropperful of the liquid into the sample well of the test cassette.
  • Wait and read: Allow the strip to develop for the specified time (commonly 5 minutes). A colored control band should appear to show the test is valid. If Group A strep antigen was present, a second line will appear at the test region. Interpretation: Two lines = positive for Strep A (test and control); one line = negative (control only).

Each step is usually illustrated in the kit manual. The entire process takes only a few minutes, so results are available long before a culture would grow.

Accuracy and Clinical Use

Rapid Strep A kits are highly specific but moderately sensitive compared to throat culture (the traditional gold standard). Most published evaluations and product inserts report sensitivities typically in the 85–95% range and specificities ~95–100%. For instance, Quidel’s QuickVue test cites ~92% sensitivity and 98% specificity; CITest’s cassette lists 95.0% sensitivity and 97.8% specificity. This means false positive results (specificity shortfall) are rare, but false negatives can occur if antigen levels are low. In practice, a positive rapid test is very reliable evidence of Group A strep infection, while a negative test does not completely rule it out.

Because of this, clinical guidelines recommend confirming negative rapid tests in certain cases. The CDC advises that in children with symptoms of strep throat, a negative rapid antigen test should be followed up by a standard throat culture. (Adults with a negative rapid test are often not cultured, as strep is less common and complications are rarer.) In short, the rapid test is fast (results in ~5 minutes) and aids prompt diagnosis and treatment, but a clinician may still confirm negatives by culture when needed.
